Abstract/Summary

The study conducted explores the toxicity levels of three herbal mouthrinses. Herbal mouthrinses are usually formulated with antimicrobial agents to inhibit the accumulation of plaque in buccal cavities. There have been reported cases of toxicity in some herbal or plant-based products, primarily due to activation of present mutagens in the cell, conversion of promutagens to mutagens, and inducing synergistic activity of the mutagens (Sarkar, et al., 1996). It is possible that usage of these products may result into cytotoxic and genotoxic damage in human oral fibroblasts. Human oral fibroblasts were cultured and then exposed to three herbal mouthrinses, namely Healthy Mouth, Nutri Smile, and Sea Fresh, to serve as test samples in the experiment. Cytotoxicity of the mouthrinses in relation to the positive control, Bleomycin and to each other, was measured through colorimetric detection using a plate reader set at 570 nm and calculation of the cytotoxicity index and IC90 from the data obtained from colorimetric detection. Based on the study, all mouthrinses displayed significantly higher cytotoxicity than Bleomycin, with Sea Fresh being significantly more toxic the other test samples. Real-time Quantitative Polymerase Chain Reaction (qRTQPCR) was performed to determine the genotoxicity of each mouthrinse, in comparison to the untreated sample and the other test samples. The quantification of data was done through the use of Rotor-Gene 3000 software. The study yielded that Sea Fresh was the most genotoxic, followed by Nutri Smile and Healthy Mouth. Overall, all of the mouthrinses tested were highly cytotoxic and genotoxic.
