Abstract/Summary

A total of six (6) PPFM bacterial isolates were isolated from the surface water of Taal Lake, Talisay. These isolates were morphologically described using colonial, microscopic and biochemical characterization. In addition, genotypic characterization using the 16S rDNA gene sequence was done. All isolates showed varying pink pigmentation. The colonies were pinpoint in diameter when grown in methanol mineral salts (MMS) agar and were small when grown in glycerol-peptone (GPA) agar. The colonies of the PPFM bacterial isolates were round or circular shape, smooth surface, had butyrous consistency, raised elevation, entire margin and opaque optical density. Under the microscope, all PPFM bacterial isolates were shown to be Gram-negative rod shaped cells with sudanophilic bodies, called poly-ß-hydroxybutyrate (PHB) granules. A total of 34 biochemical tests were performed utilizing thirty (30) biochemical activities contained in the BBL crystal enteric/nonfermenter ID kit and four (4) laboratory prepared biochemical assays. All PPFM bacterial isolates exhibited varying biochemical activities. Moreover, all isolates were positive for catalase, urease and oxidase tests which are characteristic of genus Methylobacterium. Identification of the PPFM bacterial isolates using the 16S rDNA sequence through BLAST and phylogenetic analyses revealed the isolates to belong to Methylobacterium spp.
