Abstract (English)

Biocatalysis has gained significant attention as a greener alternative to conventional organic synthesis under mild and environmentally friendly conditions, while eliminating the use of toxic reagents and harsh reaction conditions. This study investigated the potential of carrot (Daucus carota) mediated bioreduction of dicarbonyl compounds, with ethyl acetoacetate as the model substrate in evaluating the feasibility of the model reaction and in optimizing the reaction conditions. The reaction was carried out under ambient conditions using carrots as the biocatalyst in an aqueous medium, followed by reaction work-up and characterization of the product by TLC, FTIR, and NMR analyses. Optimization study revealed that fresh, peeled carrots, increased substrate loading, and 48 hours reaction time were the most favorable conditions for product formation, resulting in a maximum yield 35.3 %. The reusability study highlighted the limitations associated with the prolonged usage of the biocatalyst. The substrate scope demonstrated the successful bioreduction of isatin (24.6 %) and benzil (51.1 %), while the bioreduction of the drug substrate, Norfloxacin, was unsuccessful, highlighting the influence of substrate structure.

Abstract (Filipino)

Ang biyokatalisis (biocatalysis) ay nakakuha ng malaking atensyon bilang isang mas kalikasan-friendly na alternatibo sa pangkalahatang organikong sintesis sa ilalim ng banayad at nakabubuti sa kapaligirang mga kondisyon, habang inaalis ang paggamit ng mga nakalalasong reipiyente (toxic reagents) at matitinding kondisyon ng reaksyon. Sinuri ng pag-aaral na ito ang potensyal ng biyoreduksyon (bioreduction) gamit ang karot (Daucus carota) para sa mga tambalang dikarbonil (dicarbonyl compounds), kung saan ang etil asetasetat (ethyl acetoacetate) ang ginamit na modelong subtrato (model substrate) sa pagtatasa ng pagiging posible ng modelong reaksyon at sa pagpapatupad ng pinakamainam na kondisyon nito. Isinagawa ang reaksyon sa ilalim ng karaniwang temperatura at presyon (ambient conditions) gamit ang karot bilang biyokatalista sa isang tubigng midyum (aqueous medium), na sinundan ng proseso ng paghihiwalay ng produkto (reaction work-up) at pagkakakilanlan nito sa pamamagitan ng pagsusuring TLC, FTIR, at NMR. Ipinakita ng pag-aaral sa optimisasyon na ang mga sariwa at nabalatang karot, mas mataas na dami ng subtrato (substrate loading), at 48 oras na oras ng reaksyon ang pinakapaborableng mga kondisyon para sa pagbuo ng produkto, na nagbunga ng pinakamataas na ani (maximum yield) na 35.3 %. Binigyang-diin naman ng pag-aaral sa muling paggamit (reusability study) ang mga limitasyong nauugnay sa matagalang paggamit ng biyokatalista.
