Abstract/Summary

The responses to in vitro culture of rhizome explants of Kaempferia galanga L. from five provenances were investigated. The rhizome explants from Bicol and Cavite provenances ranked first and second, respectively both for viability and shoot formation whereas provenances showed non-specific callus induction to only three of the ten treatments tested. Comparison of mean percent viability in rhizome explants cultured under the ten different plant growth regulator treatment showed that the high level of (6.66 uM) BA and a combination of cytokinin (1.11 uM BA) and auxin (0.5 uM NAA) in were effective in promoting rhizome viability whereas a combination of cytokinin (1.11 uM BA) and auxin (3.75 uM 2,4-D) produced inhibitory effect on rhizome viability compared with the control medium. All treatments induced shoot formation in all provenances compared to control medium only but in MS + 6.66 uM BA, MS + 7.77 uM BA, MS + 1.11 uM BA + 7.0 uM K, and MS + 4.44 uM BA + 1.0 uM NAA showed almost three-fold increase implying that shoot multiplication appear to be influenced strongly by cytokinin (BA) alone or in combination with another cytokinin (K) and a combination of cytokinin (BA) and auxin (NAA). Callus was induced with the combination of low level of cytokinin (BA) and high level of auxins (NAA and 2,4-D) only Bicol, Cavite and Laguna provenances whereas the two provenances failed for the growth of callus formation. Regenerated plants were established through in vitro and ex vitro cultures from the five provenances. The differences in the response of five provenances maybe attributed to genotypic variation.
