Abstract/Summary
The chloroplast DNA gene matK, and intergenic spacer trnH-psbA, were PCR amplified from 3 selected Colocasia species (C. affinis Schott., C. gigantea (Blume) Hook.f. and C. esculenta (L.) Schott.). Using DnaSP (Rozas et al., 2003), sequence diversity of matK and trnH-psbA were evaluated. The results revealed that the sequence of trnH-psbA is more diverse in terms of polymorphic sites than matK among C. affinis, C. gigantea and C. esculenta, making it a good DNA barcode. The tree generated using matK sequences, applying Kimura 2 P and UPGMA analyses, support the current infrageneric classification of Colocasia wherein C. affinis and C. esculenta belong to Section Tuberosae, while C. gigantea belongs to Section Caulescentes (Matthews, 1991). This supports the findings of Nguyen et al. (1998) that C. gigantea is most different among and even has closer affinity to Alocasia based on mitochondrial DNA and esterase isozymes.The tree generated using trnH-psbA sequences suggest that C. gigantea and C. esculenta are most related. Neither trnH-psbA nor matK sequences support the biogeographical data provided by the World Checklist of Selected Plant Families (2009).
