Abstract/Summary
This study was a preliminary investigation on the possible nephroprotective effects of VCO against gentamicin-induced acute renal injury in Mus musculus. The study determined whether oral treatment with VCO can reduce the nephrotoxic injury of gentamicin by assessing kidney histopathology. Also, a DPPH analysis of the VCO sample was done to identify the measurement of its antioxidant levels. Thirty (30) female ICR mice were randomly assigned to six treatment groups (n=5), namely: Sham (normal saline solution [NSS]), GEN control (NSS + gentamicin 100mg/kg), VCO 5 (5 mL/kg), VCO 10 (10 mL/kg), and the two groups (VCO 5 mL/kg + gentamicin; and VCO 10 mL/kg + gentamicin). Treatment with either NSS or VCO was done for 21 days via oral gavage. The GEN and experimental groups were given once daily with gentamicin intramuscularly from days 15-21. Twenty four (24) hours following the last dose of gentamicin, kidney excision was done under humanely death. Histopathologic examination of the kidneys was done under light microscopy using a standard numerical grading scale. Histopathologic examination revealed attenuation in the development of the renal tissue injury after treatment with VCO 5 mL/kg but not VCO 10 mL/kg. The results might have been due to alteration of the effects of VCO by the acute renal injury at a higher dose. These findings suggest that oral treatment with VCO for 21 days may not have potential nephroprotective effects in reducing renal tissue injury using this animal model.
