Abstract/Summary

At present, cinnamon is considered as an important plant because of its many uses in different fields. Due to its economic and commercial importance, and the potential discovery of a new Cinnamomum species, this study aims to determine the genetic identity of an unknown cinnamon species collected from Tawi-tawi by comparing its morphological and molecular characteristics to known species namely: C. ebaloi, C. iners, C. griffithii and C. mendozai. To achieve the objective, the Cinnamomum samples were subjected to three analyses: morphological, morphometric, and molecular analysis. The leaf morphologies and morphometrics of each species were measured and compared to be able to construct a dichotomous key and a principal component analysis plot, respectively. On the other hand, molecularly, the DNA of each species were extracted, amplified and sequenced. Their MatK gene sequences were then compared to existing sequences registered in GenBank. Furthermore, their MatK gene sequences were aligned to be able to generate a contig for the comparison of the known species with that of the unknown sample based on the mismatches and gaps in their sequences. In addition, a phylogenetic tree was constructed based on their MatK gene sequences to be able to observe their relationships molecularly. Based on the results, the unknown species exhibits closely similar morphological characteristics as that of Cinnamomum mendozai but is identified as Cinnamomum ebaloi molecularly based on similarities in their MatK gene sequences.
