Abstract/Summary

Peroxidase was isolated from two Capsicum species such as siling haba (Capsicum frutescens L.) using Tris[tris(hydroxymethyl)aminomethane]-HCI buffer extraction. Three trials were done. The protein concentration of each sample was monitored through Biuret assay. Peroxidase activity assay was monitored by the reaction of H2O2, 4-aminoantipyrine and phenol. Quinoneimine dye, the produced colored solution was examined by UV-VIS Absorption Spectrophotometry. The specific activity of peroxidase was determined both for the ripe and unripe Capsicum sample. A comparison of the peroxidase activity of the two samples was done which gives a result that ripe sample of siling labuyo and siling haba yields high specific activity of peroxidase as compared to the unripe sample. The second basis of comparison, ripe sample of siling labuyo contains higher specific activity than that ripe sample of siling haba. The ripe sample of siling haba obtained lesser protein concentration than that of the ripe sample of siling labuyo. Thus, with siling labuyo's high specific activity, peroxidase can be an alternative source of medicine in diagnosing patients. Accordingly, peroxidase are capable of removing phenols which are pollutants enclosed in industrial wastewaters through enzyme-catalyzed polymerization.
